STUDIES ON THE TOXICITY AND PHARMACOLOGY OF NICOTINIC ACID

1. Acute toxicity (L.D. 50) of nicotinic acid in mice and rats ranges from 4 to 5 grams per kilogram after subcutaneous and from 5 to 7 grams per kilogram after oral administration. Nicotinic amide is about twice as toxic.

2. Prolonged oral administration of large amounts (up to 2 grams per kilogram daily) of sodium nicotinate to rats, chickens and dogs over periods up to 2 months failed to produce toxic symptoms or pathological changes of the organs.

3. In the urine collected during 24 hours after oral administration of sodium nicotinate 30 per cent of the dose could be recovered as nicotinic acid.

4. The metabolism and the circulatory and respiratory system of normal animals was not influenced by sodium nicotinate or nicotinic amide.
