CL 65,562 (4-(agr,agr,agr-TRIFLUORO-m-TOLUIDINO)-(NICOTINIC ACID), A NOVEL "HIGH CEILING" DIURETIC

CL 65,562 is a structurally novel and highly efficacious diuretic agent capable of promoting the excretion of as much as 30% of the sodium chloride filtered at the glomerulus. It is effective in all animal Species examined (rat, rabbit, guinea pig, dog and monkey) and by all common routes of administration. CL 65,562 is effective during both metabolic acidosis and alkalosis. CL 65,562 produces a diuresis similar in magnitude to that produced by furosemide and ethacrynic acid. The renal sites of action of CL 65,562 are interpreted to be the proximal tubule and the ascending limb of the loop of Henle, but an effect in the distal convoluted tubule has not been definitely excluded.
