Phenyramidol inhibits the metabolism of bishydroxycoumarin in the mouse, rabbit and man and thus prolongs the pharmacologic effect of the anticoagulant. Phenyramidol (40 mg/kg) does not affect the metabolism of meperidine, aminopyrine, zoxazolamine, pentobarbital or sulfanilamide, although it potentiates the action of pentobarbital in the SW mouse. Evidence is presented to illustrate the role of genetic factors in the metabolism of bishydroxycoumarin and antipyrine in rabbits. The implications of such findings in drug evaluation and in clinical drug therapy are discussed
