The rat, like man, has only limited capacity for the synthesis of salicylurate from salicylate and glycine. The administration of glycine has no effect on the metabolism of salicylate in man and rats or on the toxicity of salicylate in rats. Pharmacokinetic constants for salicylurate formation are in good agreement with apparent in vivo Km and Vmax values obtained from appropriate Lineweaver-Burk plots. In vivo Km values are the same in man and rats, but Vmax is about 4 times higher in man, on a body weight basis, than in rats.
