Desmethylimipramine (DMI) inhibits uptake of dl-norepinephrine-7-H3 in cat heart slices and in the mouse heart in vivo, but does not deplete previously established stores of labeled norepinephrine in the mouse heart. Although the dose of reserpine that is equivalent to 10 mg of DMI per kg in its effect on uptake is small (0.1 mg/kg), reserpine at this level retains its ability to deplete. The depleting effects of small doses of reserpine are inhibited by DMI. These and other observations suggest that DMI acts at a site in the membrane which is probably a part of the system for the uptake of norepinephrine and which has some affinity for a variety of amines.
