ESTIMATION OF THE ACETYLCHOLINE CONTENT OF RAT BRAIN BY GAS CHROMATOGRAPHY

William B. Stavinoha 1 and Leonard C. Ryan 1

1 Civil Aeromedical Research Institute, Federal Aviation Agency, Pharmacology-Biochemistry Laboratory, Oklahoma City, Oklahoma

A gas chromatographic method has been adapted for the analysis of acetylcholine in rat brain. Rat brains were frozen, extracted in acid ethanol, flash evaporated, taken up in water, reacted with borohydride and the resulting ethanol estimated on a gas chromatograph. The compounds considered possible interfering substances in the bioassay do not interfere with the estimation of acetylcholine by the chemical method. Acetylcholine levels measured by the chemical method and by bioassay are similar.
