Choline-methyl-C14 was used to label the quaternary amine stores of rat phrenic nerve-diaphragm preparations, in vitro. The efflux of Ch and ACh was then studied by analyzing the bath solutions.
The labeled Ch and ACh were precipitated with ammonium reineckate and separated by suspending the precipitates in aqueous ethanol. Paper chromatography may be used to check the methods. A mathematical method, permitting each TLQA washout curve to be self controlled in the study of drug effects was programmed for an IBM 1620 computer together with extensive statistical analysis.
The method of assay appears to be equivalent in sensitivity and precision to bioassay methods.
