STUDIES ON THE ENZYMATIC N-AND O-DEMETHYLATION OF NARCOTIC ANALGESICS AND EVIDENCE FOR THE FORMATION OF CODEINE FROM MORPHINE IN RATS AND DOGS

Christian Elison 1 and Henry W. Elliott 1

1 Department of Pharmacology and Experimental Therapeutics, University of California, San Francisco Medical Center, California

Graphical analysis of the kinetics of demethylations of codeine and dextromethorphan suggests that there exist in the rat liver different enzymes which are involved in the N- and O-demethylations of these drugs. Experimental evidence is presented for the formation of codeine from morphine in vito and in vitro.
