The distribution and fate of tritium-labeled cotinine have been studied in the mouse after intravenous injection. The studies indicate a rather even distribution among various tissues, with the exception of high concentration in excretory organs, such as the kidneys and urinary bladder, and a concentration to some extent in the cerebellum. An examination of the urine revealed the presence of the metabolites demethylcotinine, hydroxycotinine and 3-pyridylacetic acid. The metabolism of cotinine thus follows in the mouse a pattern previously established in other species. The differences in distribution between nicotine and cotinine, after intravenous administration, is discussed in terms of a possible relationship to the marked pharmacological activity of nicotine and contrasting lesser activity of its metabolite, cotinine.
