ACTION OF PHENTOLAMINE ON RAT ADIPOSE TISSUE

Phentolamine reduced the weight gain of rat epididymal adipose tissue and this effect was dose-dependent. Epinephrine and tolazoline evoked similar effects; on a molar basis the latter was about one-fifth and the former five times as potent as phentolamine. Phentolamine also depressed the content of lipid in adipose tissue depots and carcass. In carcass the concentration of saponifiable lipids was decreased and nonsaponifiable lipids increased. The lipolytic effects of phentolamine did not result from a reduction in food intake in the treated animals. Extirpation of the adrenals to remove chromaffin tissue, and institution of replacement therapy with hydrocortisone, was not effective in preventing the phentolamine effect on epididymal fat. Hypophysectomized rats also responded to the lipolytic effects of phentolamine on epididymal adipose tissue.
