The metabolism of nicotine was studied using a rabbit liver supernatant preparation with TPNH. Two-directional chromatography and radioautography revealed 8 major nicotine metabolites. Four of these had the satire mobilities as nornicotine, demethylcotinine and pyridylacetic acid standards. The other four have not been identified. Nornicotine, cotinine and demethylcotinine have been isolated and identified by physical and chemical methods. The oxidative and demethylation processes in the metabolism of nicotine are discussed.
