THE EFFECT OF PHENOTHIAZINE DERIVATIVES ON THE DISAPPEARANCE OF SULFOBROMOPHTHALEIN FROM MOUSE PLASMA

The plasma dye-disappearance curve of sulfobromophthalein (150 mg/kg, i.v.) has been shown to be altered by pretreating mice with various phenothiazine derivatives. Dose-response studies, the duration of action, and the effect of chronic treatment were investigated, using orally administered chlorpromazine, thioridazine or mercaptothiazinyl. No residual hepatic alteration in function was observed after a 14-day treatment period. However, tolerance was found to develop to the dye retention phenomenon after 14 days of treatment. It is suggested that the observed plasma dye retention is due to a decrease in hepatic blood flow.
