6-AMINONICOTINAMIDE, A CENTRAL NERVOUS SYSTEM DEPRESSANT

6-Aminonicotinamide, a nicotinamide antimetabolite, was found to possess CNS depressant activity. The agent reduced the spontaneous activity of mice, potentiated ethanol and barbiturate narcosis and had protective action against picrotoxin- and pentylenetetrazol-induced convulsions. The rate of oxidation of ethanol was slightly reduced in mice pretreated with the agent. The rate of disappearance of thiopental in the total mouse was increased during the first 30 minutes but reduced below control values in the 30- to 60-minute time period.

Assay of pyridine nucleotides revealed moderate but statistically significant reduction of TPNH and DPNH concentration in liver tissue of 6-aminonicotinamide treated animals. DPN values in brain homogenates decreased slightly with increasing dose. Possible mechanisms of action were discussed.
