BIOCHEMICAL AND ELECTRON MICROSCOPIC STUDY OF PARTICLES ISOLATED FROM MASTOCYTOMA CELLS

Paul Hagen 1, Russell J. Barrnett 1, and Fu-Li Lee 1

1 Departments of Pharmacology and Anatomy, Harvard Medical School, Boston, Massachusetts

Morphological studies of mastocytoma cells have been combined with centrifugation techniques and biochemical and bioassay procedures to determine the intracellular sites of storage of histamine, 5-hydroxytryptamine and heparin.

Histamine, heparin and 5-hydroxytryptamine are contained within specific granules, which are clearly separable from mitochondria and other intracellular organelles.

Electron microscopie observations of the ultrastructure of mastocytoma cells and of the isolated particles are reported.
