METABOLIC ASPECTS OF THE POSITIVE INOTROPIC ACTION OF FLUORIDE ON RAT VENTRICLE

Jerome M. Covin 1 and David A. Berman 1

1 Department of Pharmacology, School of Medicine, University of Southern California, Los Angeles, California

The effect of substrates on the inotropic action of fluoride on the electrically stimulated rat ventricle strip was investigated. Sodium fluoride exerted a marked positive inotropic effect in pyruvate, glucose and substrate-free media and a negative inotropic effect in an acetate medium. Experiments performed under conditions of simultaneous recording of oxygen consumption and force of contraction of electrically stimulated rat ventricle strips demonstrated that the positive inotropic effect of fluoride was not accompanied by a significant increase in the oxygen consumption. The effect of frequency of stimulation on the response of the ventricle strip to fluoride was investigated; it was found that the magnitude and duration of the positive inotropic effect was inversely related to the frequency of stimulation.
