Narcotine hydrochloride produces systemic vasodepression which is incompletely blocked by diphenhydramine but is unaffected by atropinization, mid-cervical vagotomy and/or carotid sinus and body denervation.
It also increases coronary blood flow and femoral blood flow. Coronary blood flow is increased without significant effects on the other cardiovascular variables.
Bronchoconstriction is produced in the dog.
Tachyphylaxis develops to the vascular and bronchial effects of this compound. In addition, cross-tachyphylaxis develops between marcotine and 48/80.
It is concluded that tue mechanism for these changes is due to histamine liberation.
