Prolonged hypotension occurs after the intravenous administration of 2, 2'-oxybis (p-phenylenemethylene) bis-isothiourea (BITU). Brief hypertension precedes the hypotension.
The hypertension is accompanied by reflex vasoconstriction and the hypotension by brief reflex vasodilatation in the perfused hind limb of of the dog. Reflex vasoconstriction is probably due to ganglionic effects; reflex vasodilatation is due, at least in part, to an action in the head. A more persistent vasodilator action is also seen and is attributed to a direct effect on the vasculature.
Cardiac output is not reduced in the dog heartlung preparation by large doses of BITU but responses to increased right atrial pressure are impaired. In the intact animal, the electrocardiogram remains normal until just before death.
BITU potentiates the actions of norepinephrine and DMP (P1, 1'-dimethyl-4-phenylpiperazinium).
A rise in plasma histamine levels was not detected in dogs or cats which had received doses of BITU large enough to cause marked hypotension. Following lethal or near lethal doses, plasma levels of histamine were increased.
