ANALGESIC-POTENTIATING AND DIURETIC EFFECTS OF 1-DIMETHYLAMINO-3-CYANO-3-PHENYL-4-METHYL-HEXANE HCl (Z-4) AND 1-DIMETHYLAMINO-2-PHENYL-3-METHYL-PENTANE HCl (Z-134)

Simple and satisfactory methods for assaying analgesic and diuretic agents in rats are described.

1-Dimethlamino-2-phenyl-3-methyl-pentane HCL (Z-134) and to a lesser extent 1-dimethylamino-3-cyano-3-phenyl-4-methyl-hexane HCL (Z-4) show striking analgesic-potentiating efficacies by different methods in different species of animals, compared with chlorpromazine.

Intramuscular Z-134 and oral Z-4 exhibit remarkable diuretic activity in rats and compare favorably with Mercuhydrin and Diamox.

In non-toxic doses, the Z compounds produce prolonged respiratory stimulant and intestinal relaxant effects in dogs.

Moderate local and topical anesthetic effects were also demonstrated with these new drugs in animal studies.

Acute toxicity studies in animals indicate that these drugs are relatively non-toxic.
