CENTRAL DEPRESSANT AND ANTICONVULSANT ACTIVITY OF COMPOUNDS ISOMERIC WITH 2-METHYL-2-n-PROPYL-1 ,3-PROPANEDIOL DICARBAMATE (MILTOWN)

Nine compounds isomeric with 2-methyl-2-n-propyl-1, 3-propanediol dicarbamate (Miltown) were tested in mice for their ability to produce paralysis, antagonize convulsions produced by pentylenetetrazole and modify maximal electroshock seizures. Miltown was the most potent compound in the series in paralyzing action and in preventing convulsions and death caused by pentylenetetrazole. The dicarbamates of 2-n-butyl-1, 3-propanediol and of 2-isobutyl-1, 3- propanediol were outstanding in their ability to modify maximal electroshock seizures.

The relation between pharmacological action and physical properties is briefly discussed. The paralyzing potency and the ability of compounds to protect from pentylenetetrazole convulsions correlate well with the melting points but there was no significant correlation between the melting points and the ability of compounds to modify electroshock seizures.
