The effects of fluoroacetate on dog renal slices were studied. It was shown that oxygen consumption of these slices is not markedly changed while the PAH concentrating ability is markedly depressed.
The concentration of calcium, potassium and a number of Krebs cycle intermediates in the buffer solution did not change the NaFAc effects on PAH uptake. Acetate, pyruvate, butyrate, valerate and monoacetin when added to the buffer markedly protected the PAH concentrating mechanism against NaFAc. Lactate did not protect. Increasing the pH of the buffer reduced the action of NaFAc. The results obtained are discussed in the light of Peters' theory of the action of fluoroacetate.
