It has been demonstrated beyond reasonable doubt that the initial step in the origin of streptomycin dependence in Escherichia coli is a mutation that occurs in the complete absence of streptomycin. These mutants must undergo further changes of a mutational or adaptive nature in the presence of streptomycin, before they can give rise to streptomycin-dependent colonies.
