The coronary dilator action of six antihistamine compounds was studied quantitatively in the intact dog. All of the compounds appeared to have some activity and are in the following descending order of potency: RP-3015, Phenergan, Diphenhydramine, Diparcol, Neoantergan, Pyribenzamine. There appeared to be no direct relationship between coronary dilator activity and antihistamine activity. The coronary dilatation following intracoronary injection occurred without any consistent change in blood pressure, heart rate, stroke volume, cardiac output, total peripheral resistance, or cardiac work. Coronary resistance was always reduced. RP-3015 was found to be effective after intravenous injection.
