A number of compounds related to 1-n-butylamino-3-p-toluidino-2-propanol (W 181) have been investigated for their convulsant activity and awakening action in paralysis caused by mephenesin.
1-Alkylamino-3-p-toluidino-2-propanols possessed good awakening action and were potent convulsants provided that the alkyl group was 3 to 5 carbon atoms in length.
Lack of a methyl group or a shift in the position of the methyl group to the ortho or meta position in the aniline nucleus resulted in loss of analeptic action.
None of the convulsants effective in shortening mephenesin paralysis possessed an appreciable margin between convulsant and lethal doses.
