The intracellular distribution of histamine was studied in dog liver by means of differential centrifugation to isolate the cellular fractions and a chemical method for histamine estimation. The mitochondrial fraction isolated from dog liver contained the major portion of the histamine present in the whole homogenate. Anaphylactic shock in the dog resulted in most instances in a significant release of histamine from the mitochondrial fraction with a variable loss from the other subcellular fraction. The relation of histamine distribution to the intracellular localization of antibody formation, antigen binding and antigen-antibody reaction was discussed.
