A new method is described for the histochemical localization of acetylcholinesterase in fresh frozen sections of nervous tissue, based upon the acetyl-esterase function of this enzyme. Evidence by means of specific inhibitors is given that staining in nervous tissue is due mainly to acetylcholinesterase activity. Enzymatic activity was demonstrated in the cytoplasm and cell membranes of neurocytes, in the neuropile and in myoneural junctions. Cell nuclei were free of enzymatic activity. Illustrations are given.
