The guinea pig ileum has been investigated as a method for assaying Ach. It has the advantage of great rapidity and ease of operation. Statistical analysis has shown that it will give accurate and reproducible results.
A method has been devised for separating Ach from various alkaloids including eserine, morphine, atropine, hysocine, and strychnine. It may also be separated from epinephrine and histamine, by the same technique. The method involves precipitation as a double choline-acetylcholine reineckate. The results of a number of precipitations have been analysed statistically, and none are significantly different from the theoretical yield.
The method is particularly applicable to the assay of Ach synthesised by means of choline acetylase in acetone-dried brain powders.
