QUINIDINE-LIKE ACTIVITY OF THEPHORIN

William Schallek 1

1 Department of Pharmacology, Hoffmann-La Roche Inc., Nutley 10, New Jersey

Thephorin has been found to be more active than quinidine in the following tests: (1) refractory period of isolated rabbit auricles; (2) electrically-induced auricular fibrillation in the cat; (3) acetylcholine-induced auricular fibrillation in the dog; (4) epinephrine-induced ventricular premature contractions in the dog.

When the effects of these drugs were compared on the electrocardiogram of the cat, Thephorin was found to cause greater prolongation of the refractory period than quinidine and less slowing of conduction.

The toxicity of Thephorin in mice is greater than that of quinidine.
