Under aerobic conditions, incubation of rat liver slices with Krebs-Ringerbicarbonate to which codeine has been added, results in the disappearance of codeine and the formation of morphine. The liberated morphine is further metabolized by conjugation.
The total amount of morphine formed represents less than one-half the metabolized codeine.
The disappearance of codeine is markedly reduced by the presence of M/1500 mono-iodoacetic acid; the presence of M/50 NaF has no apparent effect on the disappearance of codeine.
A rapid method for the assay of codeine concentration is presented.
