The effect of pentobarbital sodium on a variety of enzyme systems has been studied. The barbiturate is shown to block the pyruvic acid oxidase system, most likely at the oxidation of the two-carbon fragment. The barbiturate does not block the oxidation of flavoprotein by either oxygen or cytochrome c. Rather, the essential sulfhydril groups of the dehydrogenase of the pyruvic acid system are shown to be the barbiturate-sensitive step. The relation of this enzyme inhibition to narcosis in vivo is discussed.
