INHIBITION OF THE KIDNEY SUCCINIC DEHYDROGENASE SYSTEM BY MERCURIAL DIURETICS

Mercurial diuretics when administered intravenously reversibly inhibit the succinic dehydrogenase system of dog and rat kidney cortices. Maximal inhibition occurs in the dog kidney during peak mercurial diuresis and recovery of enzyme activity is coincidental with recovery from diuresis. The administration of 2,3-dimercaptopropanol prior to removal of the kidney completely abolishes the effect of the mercurial diuretic on the enzyme system. When compared with p-chloromercuribenzoate, a potent inhibitor of enzymes containing sulfhydryl groups essential to activity, mercurial diuretics are approximately one-half as active for producing inhibition of the succinic dehydrogenase system.
